Dexamethasone dose contained by mice?
I need to give mice 5mg/kg of Dexamethasone in 100uL saline. The average counterweight of the mice is 24 grams. The concentrations of Dexamethasone stock (water soluble powder) is 65 mg/g.
I figured that we need 0.00184g (of the stock solution). Is this right? What about the 100uL saline though?
Answers:
First the individual dose:
24gm = 0.024kg
0.024kg * 5mg/kg = 0.12mg per dose
Each dose is 100ul or 0.1ml so respectively injection will be 0.12mg/0.1ml
Next make a stock injection solution with a conc. of 0.12mg/0.1ml
I'll make 5ml (50 doses) as an example.
How much dex. total?
0.12mg/dose * 50 doses = 6mg total dex. needed
How much stock powder? (65mg dex. per 1gm powder)
65mg/1gm = 6mg/xgm
x = (1gm*6mg)/65mg
x = 0.092gm or 92mg powder
Make the injection solution:
Add a sufficient mass of saline to 92mg of powder to make a total volume of 5ml.
Each 100ul will contain 0.12mg Source(s): Pharmacist
